Overview
Shiitake (Lentinula edodes) is an edible mushroom native to East Asia, cultivated for centuries in Japan, China, and Korea on hardwood logs. In Poland it's sold almost exclusively under its Japanese trade name 'shiitake' — fresh and dried, as well as standardized capsule extracts.
Shiitake is among the most intensively studied functional mushrooms in the world, largely thanks to lentinan — a beta-1,3/1,6-glucan polysaccharide that in Japan is a registered drug administered intravenously as adjunct oncology therapy. Separate from this clinical form, oral consumption of the fruiting body and standardized extracts has been studied for effects on cellular immunity and lipid profile in healthy and partly metabolically burdened adults, with results pointing to modest but reproducible effects.
From a practical standpoint, shiitake will most realistically help healthy people looking for gentle, long-term dietary immune support — daily consumption of dried fruiting bodies for 4 weeks improved selected cellular and mucosal immunity parameters in healthy young adults. Similarly, people with borderline elevated cholesterol can expect modest, complementary support for their lipid profile — but only as an addition to, not a replacement for, standard treatment. Shiitake, however, will not help people expecting the therapeutic effect known from injectable lentinan used in oncology — that's an entirely different form, dose, and route of administration than the dried fruiting bodies or capsules available to consumers.
Where it's found
Shiitake's natural habitat is the trunks of dead and dying hardwood trees (oak, chestnut, beech) in the humid forests of East Asia — Japan, China, and Korea. Today the mushroom is grown commercially worldwide, traditionally on hardwood logs, and industrially on sawdust substrates.
History of use
Documented shiitake cultivation in China dates back to the Song dynasty (roughly the 13th century), and in Japan the traditional log-cultivation method ('hodakagi') developed during the Edo period. For centuries the mushroom has been a staple of Chinese and Japanese cuisine and medicine, valued as natural immune support.
Mechanism of action
The main studied component is lentinan — a large-molecule beta-glucan that stimulates macrophages, NK cells, and T and B lymphocytes through interaction with pattern-recognition receptors (including dectin-1). In human studies, daily consumption of fruiting bodies increased the proliferation and effector capacity of γδ-T and NK-T lymphocytes and raised secretory IgA levels. A second important mechanism is the action of eritadenine — a compound present in fruiting bodies that affects methionine and cholesterol metabolism, partly explaining the drop in triglycerides observed in clinical studies.
At the molecular level, lentinan is recognized primarily by the dectin-1 receptor (CLEC7A) on the surface of macrophages and dendritic cells, partly with the involvement of the TLR-2 receptor as well — this combination initiates a signaling cascade leading to activation of the transcription factor NF-κB. Eritadenine, meanwhile, works through a different pathway — as an adenine nucleoside analog it inhibits S-adenosylhomocysteine (SAH) hydrolase activity, disrupting the methionine methylation cycle, which in turn changes the ratio of phosphatidylcholine to other phospholipids in the liver, indirectly affecting the rate at which cholesterol and LDL are cleared from circulation.
Lentinan binding to the dectin-1 receptor
Lentinan's beta-1,3/1,6-glucan is recognized by the dectin-1 receptor (and partly TLR-2) on macrophages and dendritic cells.
Activation of NK-T cells and γδ-T lymphocytes
Signaling through NF-κB leads to increased proliferation and effector capacity of NK-T cells and γδ-T lymphocytes.
Rise in sIgA and drop in CRP
The end result of cellular activation is a rise in secretory IgA and a drop in CRP after about 4 weeks of daily consumption.
Eritadenine and lipid metabolism
In parallel, eritadenine inhibits SAH hydrolase, altering hepatic phospholipid metabolism and supporting cholesterol clearance from circulation.
