Overview

Enokitake (Flammulina velutipes), also called Velvet Foot in English-speaking countries, is an edible mushroom that grows wild on the trunks of deciduous trees across Asia, Europe and North America. The form sold in stores looks nothing like the wild mushroom: grown industrially in darkness and under elevated CO2, it forms long, thin, white stems with tiny caps. In Asian cuisine it goes mainly into soups and hot-pot dishes.

Enokitake is often brought up in a health context mainly because of FIP-fve — an immunomodulatory protein studied for its effect on T lymphocytes. The popular anecdote about a Japanese farming village with lower cancer rates rests on a single, methodologically limited observational study. Enokitake remains primarily a food product, not a standalone supplement.

Enokitake will realistically help mostly as a low-calorie, fiber-rich addition to the diet — on that front the benefit is certain and applies to anyone who cooks it properly. People counting on a measurable immunomodulatory effect from FIP-fve are unlikely to see it in practice: the protein has been studied only in isolation and in vitro, not after eating the fruiting bodies as food. For pregnant women, older adults and people with weakened immunity, enokitake can actually be harmful if it ends up on the plate raw or undercooked.

Where it's found

Grows wild on trunks and dead wood of deciduous trees (including elm, mulberry, persimmon) across Asia, Europe and North America, typically in autumn and winter. The form sold in stores is grown industrially in darkness and under elevated CO2 concentration, producing long, pale stems that look completely different from the wild mushroom.

History of use

In Japanese, Chinese and Korean cuisine, enokitake has long been used, primarily in soups and hot-pot dishes (e.g. nabemono), where thin bunches of the mushroom are added a few minutes before serving.

Mechanism of action

The best-described component is FIP-fve — a protein belonging to the family of fungal immunomodulatory proteins. In vitro studies show it acts as a mitogen on human lymphocytes, activates T lymphocytes and increases interferon-gamma secretion via the p38 MAPK pathway. No study to date confirms an analogous effect from simply eating the fruiting bodies as food.

The p38 MAPK pathway activated by FIP-fve is one of three main mitogen-activated kinases (alongside ERK and JNK) — its stimulation leads to phosphorylation of transcription factors and increased expression of the IFN-gamma gene in T lymphocytes. Key caveat: in both cited studies the effect was shown on purified protein under in vitro conditions — the content and bioavailability of FIP-fve in cooked fruiting bodies eaten as food remains unstudied.
