Overview
NMN (nicotinamide mononucleotide) is a precursor in the NAD+ (nicotinamide adenine dinucleotide) synthesis pathway — a coenzyme present in every cell of the body and essential for hundreds of metabolic reactions, including mitochondrial energy production and DNA repair. NAD+ levels naturally decline with age — in people 60 and older they can be as much as half those of young adults, which some researchers link to the general drop in metabolic fitness that accompanies aging.
NMN, alongside the related compound NR (nicotinamide riboside), became one of the most talked-about topics in the longevity world following publications by researcher Shin-ichiro Imai of Washington University and popularization by well-known aging researchers such as David Sinclair of Harvard. This drove a rapid surge in market popularity that has significantly outpaced the pace of solid, large-scale human clinical trials.
At this stage of the research, it's hard to point to a group that can confidently expect to 'feel' an NMN effect in the sense of a subjectively noticeable improvement — the available human clinical data are few, short-term, and focused on narrow, objectively measured parameters (for example, muscle insulin sensitivity in women with prediabetes in a 10-week study), rather than subjective well-being or energy. Realistically, NMN makes sense only for people deliberately experimenting at an early stage of the evidence, who accept the high cost and the lack of any guaranteed effect — it is not a first-choice supplement for people starting out in biohacking, nor for anyone expecting a fast, measurable difference. History of use

Mechanism of action
NMN is absorbed and converted in the body into NAD+ via a two-step metabolic pathway (the so-called salvage pathway). The research hypothesis — still actively being tested, not yet conclusively confirmed — holds that raising the NAD+ levels that decline with age, via precursor supplementation such as NMN, might slow certain aspects of cellular aging.
The enzyme NAMPT is the natural, rate-limiting step of endogenous NMN synthesis from nicotinamide in the body — its activity also declines with age, which is one of the reasons NAD+ levels fall during aging. After oral administration, NMN must be absorbed and delivered into cells — in mouse studies, a dedicated membrane transporter, Slc12a8, was identified as responsible for direct cellular uptake of NMN in the intestine, though its role in humans is still under investigation. Elevated NAD+ is a key substrate for sirtuins, particularly SIRT1 and SIRT3, enzymes that regulate inflammation, mitochondrial function and the response to metabolic stress — it is their activity in animal models that has been linked to the observed lifespan extension, though translating this mechanism to humans remains a hypothesis.
